TRANSIL Assays

For protein binding application our chemists prime the bead surface so that it can covalently bind proteins. The resulting protein-coated beads have very little exposed silica surface which practically eliminates any contact of the silica with the solution and keeps unspecific binding of drug candidates or other test substances minimal.

For membrane permeability applications the beads have lipid bilayer membranes attached via non-covalent interactions. This mimics the fluidity of native membranes, and allows the complete range of complex interactions that occur between a drug candidate, or other test substance, and the membrane molecules.

Kit Contents

Protein Binding and SA/AGP kits :The kit consists of ready-to-use 96 well microtiter plates. One plate can be used for measuring up to 12 compounds.

The assay requires only 5 steps: (i) addition of drug candidate, (ii) mixing and incubation for 12 minutes, (iii) removal of beads by centrifugation, (iv) sampling of supernatant, and (v) quantification of drug candidate.

High Sensitivity Binding kits :  

The kit consists of silanized glass vials pre-filled with buffer and membrane beads.

The assay requires only 6 steps: (i) addition of plasma, (ii) addition of drug candidate, (iii) incubation for 30 minutes while mixing, (iv) removal of beads by centrifugation, (v) precipitation of plasma, and (vi) quantification of compounds.

TRANSIL Workflow

1. Prepare compound (5 minutes)
2. Add compound
3. Mix
4. Incubate and mix (12 minutes)
5. Centrifuge (10 minutes)
6. Transfer supernatant
7. Quantify drug in supernatant
8. Paste data into spreadsheet for analysis
